Let's add a little devil's advocacy to the issue of respiratory uptake (colds, infections, etc), or that shots continue to rule the domain.
1. What if you pick an injection site that is already so scarred from previous injections that it slows or limits the timely uptake from the shot?
2. What if you mis-fire your pen prematurely, or don't load the proper amount into the syringe to begin with? At my age 3, 5, 6, and 8 are hard to distinguish. Couldn't this lead to improper dosing?
3. If the lungs fail to function with a cold, wouldn't most people suffocate for lack of oxygen? It is, after all arriving the same way the drug is, right?
4. Do asthmatics have to find alternative remedies for their inhalers when they have a cold?
Perhaps we don't give our lungs enough credit?!
Just some thoughts to counter any suggestion that the status quo is any better than the proposed new technology.
To the point of mis-dosing, it sounds like the kinetics of Afrezza are going to be on its side, offering much more lenient dosing requirements, mainly due to the fast peak concentrations, and minimal time to leave the body. This bodes much better for an ultra fast acting drug than one that could take hours to leave your system.
I think the folks raising these concerns are not applying all the knowledge and information to answer the question, but relying on old school understanding of the dynamics and risks with current insulins.
All just my opinion, of course.
