Yup. Preclinical. Different biodistribution, pharmacokinetic characteristics, downstream effects, etc.
And from what i understand JQ1 does cross the blood brain barrier, where Apabetalone probably (?) does not..
So that and other major differences could mean completely different results... or .. or not? (maybe?)
I guess we won't know unless we look. I was just sort of hoping the parallels in the portions of the mechanisms discussed made it WORTH taking a look. Your cautions are noted and well taken.
The question of whether a drug could be effective without crossing the blood brain barrier, IF it changed the right parameters that in turn affected the brain... remains sort of open (at least for me).
Your comment about the Alzheimers results seems to indicate that that could be possible, at least in certain circunstances (lipoproteins, circulating factors, etc.... ALP too) Though whether it could be beneficial for epilepsy is yet another question.
These articles also seem to suggest the possibility of some beneficial effectiveness even without crossing the blood brain barrier... though they are not about epilepsy.
Am guessing that if Apabetalone did cross the blood brain barrier, they would have figured that out and we would have heard that by now... ? So probably not?
Are there compounds that cross the blood brain barrier to a greater or lesser degree, or is it a pretty black and white thing?
The above articles don't explicitly say that Apabetalone does not, but they do sort of leave the impression that it does not.
Even so, they suggest that it does still effect the vascular epithelium in ways that could help with inflammatory damage and possibly other things (as you mentioned) that in turn could be helpful for dementia.
Wonder whether RVX has a blood brain barrier "crosser" within their hypothetical larger suite of compounds, or for that matter whether Apabetalone could work in combo with...was it BioOasis that had the one that could "carry" other compounds across the blood brain barrier?
Just rambling questions at this point.
Either way, it sure would be nice to find cures for some of these diseases.
