The BET/BRD inhibitor JQ1 improves brain plasticity in WT and APP mice by Benito et al.
Abstract: "Our findings suggest that JQ1 could be used as a therapy against dementia and should be further tested in the context of learning and memory."
Discussion: "Our results are encouraging for the development of dementia therapies or to treat age-associated memory decline and suggest that BET inhibitors should be further tested in other animal models of cognitive impairment."
Really good to see this paper, since an earlier paper from 2015 by Korb et al suggested that BET inhibitors may impair memory formation.
BET protein Brd4 activates transcription in neurons and BET inhibitor Jq1 blocks memory in mice.
Benito et al acknowledge the Korb et al study in their discussion and address the potential inconsistencies between their study and that of Korb et al by stating:
"...it is thus possible that systemic administration of JQ1 improves hippocampus-dependent memory function and synaptic plasticity, while it impairs other forms of cortical plasticity, an issue that should be addressed in future research."
