There is limited data in the public domain about apabetalone's ability to get across the BBB, as well as limited data on effects of apabetalone on cognition/CNS disease endpoints in animal/human models. Most of what is out there is studies in cells and not animals. The exception is the cognition sub-study of BETonMACE in which they saw some encouraging positive data. These effects may be due to apabetalone getting across the BBB and eliciting direct effects in the brain, or due to indirect effects via apabetalone modulating circulating factors or BBB endothelial cells. Too little info to know for sure. But if apabetalone reaching the brain is indeed limiting and important for its effects on cognition, then facilitated delivery of apabetalone by a xB3 like carrier would be expected to yield a more pronounced benefit.
